=== Pharmacokinetics === Metabolic studies indicate that protriptyline is well absorbed from the gastrointestinal tract and is rapidly sequestered in tissues. Relatively low plasma levels are found after administration, and only a small amount of unchanged drug is excreted in the urine of dogs and rabbits. Preliminary studies indicate that demethylation of the secondary amine moiety occurs to a significant extent, and that metabolic transformation takes place in the liver. It penetrates the brain rapidly in mice and rats, and moreover that which is present in the brain is almost all unchanged drug. Studies on the disposition of radioactive protriptyline in human test subjects showed significant plasma levels within 2 hours, peaking at 8 to 12 hours, then declining gradually. Urinary excretion studies in the same subjects showed significant amounts of radioactivity in 2 hours. The rate of excretion was slow. Cumulative urinary excretion during 16 days accounted for approximately 50% of the drug. The fecal route of excretion did not seem to be important. Protriptyline has uniquely low dosing among TCAs, likely due to its exceptionally long terminal half-life. It is used in dosages of 15 to 40 mg/day, whereas most other TCAs are used at dosages of 75 to 300 mg/day. The maximum dose is 60 mg/day. ==== Space disposal ==== Space disposal is attractive because it removes nuclear waste from the planet. It has significant disadvantages, such as the potential for catastrophic failure of a launch vehicle, which could spread radioactive material into the atmosphere and around the world. A high number of launches would be required because no individual rocket would be able to carry very much of the material relative to the total amount that needs to be disposed. This makes the proposal economically impractical and increases the risk of one or more launch failures. To further complicate matters, international agreements on the regulation of such a program would need to be established. == Function == The ELN gene encodes a protein that is one of the two components of elastic fibers. The encoded protein is rich in hydrophobic amino acids such as glycine and proline, which form mobile hydrophobic regions bounded by crosslinks between lysine residues. === Steroid and protein hormones === Many other regulatory systems are involved in the transition of cartilage to bone and in bone maintenance. A particularly important bone-targeted hormonal regulator is parathyroid hormone (PTH). Parathyroid hormone is a protein made by the parathyroid gland under the control of serum calcium activity. PTH also has important systemic functions, including to keep serum calcium concentrations nearly constant regardless of calcium intake. Increasing dietary calcium results in minor increases in blood calcium. However, this is not a significant mechanism supporting osteoblast bone formation, except in the condition of low dietary calcium; further, abnormally high dietary calcium raises the risk of serious health consequences not directly related to bone mass including heart attack and stroke. Intermittent PTH stimulation increases osteoblast activity, although PTH is bifunctional and mediates bone matrix degradation at higher concentrations. == History == Tom Kudirka, originally from Omaha, Nebraska, and an alumnus of Bellevue West High School, had studied business information systems and electronics before moving to Tulsa to work for Lucent Technologies as a field engineer. Thereafter, he had established multimedia companies, including one dealing with speech recognition. Because Kudirka "thought he was bored", he founded 2015 in Tulsa in May 1997. The name was said to have been chosen for an undisclosed "very significant" reason, although Kudirka later said he did not know what it meant. Kudirka recruited developers he had discovered online and had them relocate to Tulsa. The designer Zied Rieke was the first hire. Through Kudirka's connections with Ritual Entertainment and its CEO, 2015 came to develop Wages of Sin, an expansion pack for Ritual Entertainment's 1998 game Sin. It was released in February 1999. On April 3, 2000, 2015 announced the opening of its Trainwreck Studios division for budget-price games with a wider target audience. The first game therein, Laser Arena, was designed to be playable by Kudirka's three-year-old son and released in September 2000. 2015 was simultaneously working on its third game, Heavy Gear Extreme.
